Primary outcomes were verbal (Buschke Selective Reminding Test [SRT]) and visual (Brief Visual Memory Test-Revised [BVMT-R]) memory, and attention (Trail Making A) was a secondary outcome. FDDNP-PET signals (15 curcumin, 15 placebo) were determined in amygdala, hypothalamus, medial and lateral temporal, posterior cingulate, parietal, frontal, and motor (reference) regions. Mixed effects general linear models controlling for age and education, and effect sizes (ES; Cohen&#8217;s d) were estimated.<\/p>\n<p style=\"text-align: justify;\"><strong class=\"sub-title\">Results: <\/strong>SRT Consistent Long-Term Retrieval improved with curcumin (ES = 0.63, p = 0.002) but not with placebo (ES = 0.06, p = 0.8; between-group: ES = 0.68, p = 0.05). Curcumin also improved SRT Total (ES = 0.53, p = 0.002), visual memory (BVMT-R Recall: ES = 0.50, p = 0.01; BVMT-R Delay: ES = 0.51, p = 0.006), and attention (ES = 0.96, p &lt; 0.0001) compared with placebo (ES = 0.28, p = 0.1; between-group: ES = 0.67, p = 0.04). FDDNP binding decreased significantly in the amygdala with curcumin (ES = -0.41, p = 0.04) compared with placebo (ES = 0.08, p = 0.6; between-group: ES = 0.48, p = 0.07). A growing body of literature suggests that potent inhibitors of tumor necrosis factor-\u03b1 (TNF-\u03b1) have potential to improve cognitive performance.<\/p>\n<p><strong class=\"sub-title\">Objective: <\/strong>In this review, we summarize the evidence regarding the potential for TNF-\u03b1 inhibition to prevent AD and improve cognitive function in people at risk for dementia.<\/p>\n<p><strong class=\"sub-title\">Methods: <\/strong>We conducted a literature review in PubMed, screening all articles published before July 7, 2019 related to TNF blocking agents and curcumin (another TNF-\u03b1 inhibitor) in the context of AD pathology. The keywords in the search included: AD, dementia, memory, cognition, TNF-\u03b1, TNF inhibitors, etanercept, infliximab, adalimumab, golimumab, and curcumin.<\/p>\n<p><strong class=\"sub-title\">Results: <\/strong>Three large epidemiology studies reported etanercept treated patients had 60 to 70% lower odds ratio (OR) of developing AD. Two small-randomized control trials (RCTs) demonstrated an improvement in cognitive performance for AD patients treated with etanercept. Studies using animal models of dementia also reported similar findings with TNF blocking agents (etanercept, infliximab, adalimumab, Theracurmin), which appeared to improve cognition. Breast cancer was diagnosed in 2013 and she has been receiving anastrozole treatment for 6 years after local mass excision surgery and radiotherapy. <\/span><\/p>\n<p style=\"text-align: justify;\"><span>After a comprehensive geriatric evaluation, cognitive impairment due to systemic cancer therapy was detected and treatment was started with Theracurmin 90 mg twice a day therapy. Theracurmin is more bioavailable form of curcumin, generated with submicron colloidal dispersion. <\/span><\/p>\n<p style=\"text-align: justify;\"><span>Mice were treated with Theracurmin (100, 300 and 1,000 mg\/kg) for 12 weeks and were subjected to the novel object recognition test and the Barnes maze test. Theracurmin-treated mice showed significant amelioration in recognition and spatial memories compared those of the vehicle-treated controls. In addition, the antioxidant activities of Theracurmin were investigated by measuring the superoxide dismutase (SOD) activity, malondialdehyde (MDA) and glutathione (GSH) levels. The increased MDA level and decreased SOD and GSH levels in the vehicle-treated 5XFAD mice were significantly reversed by the administration of Theracurmin. <\/span><\/p>\n<p style=\"text-align: justify;\"><span>Moreover, e<\/span><span>e observed that Theracurmin administration elevated the expression levels of synaptic components, including synaptophysin and post synaptic density protein 95, and decreased the expression levels of ionized calcium-binding adapter molecule 1 (Iba-1), a marker of activated microglia. <\/span><\/p>\n<p style=\"text-align: justify;\"><span>These results suggest that Theracurmin ameliorates cognitive function by increasing the expression of synaptic components and by preventing neuronal cell damage from oxidative stress or from the activation of microglia. However, curcumin shows poor oral bioavailability. <\/span><\/p>\n<p style=\"text-align: justify;\"><span>The purpose of this study was to investigate the protective effects of highly bioavailable curcumin, Theracurmin(\u00ae), and curcumin, against sodium nitroprusside (SNP)-induced oxidative damage in mice brain. Intrastriatal microinjection of Theracurmin(\u00ae) or curcumin with SNP significantly protected against SNP-induced brain damage and motor dysfunction.<\/span><\/p>\n<p style=\"text-align: justify;\"><span> Oral administration of Theracurmin(\u00ae) (1 and 3 g kg(-1), containing 100 and 300 mg kg(-1) curcumin, respectively) significantly protected against SNP-induced brain damage and motor dysfunction. However, oral administration of 300 mg kg(-1) curcumin did not protect against motor dysfunction induced by SNP. <\/span><\/p>\n<p style=\"text-align: justify;\"><span>These results suggest that curcumin and Theracurmin(\u00ae) have protective effects against SNP-induced oxidative damage.
